Just got X4, and the Timelines missions are incredibly frustrating and really bad for space-sim fare.

Do I have to do them before going into Open universe? It looks like they unlock content in open universe?

You can ignore them. They do unlock some a few sectors and ships in the open universe, but you don't have to complete Timelines first. You can play in your open universe, and do Timelines later, and they will unlock in your existing game.

You don't have to do them, and can skip them. The game insisting that you play Timelines after tutorials was an awful design decision that hasn't been fixed.

Yes, they do unlock things in open universe.

Once you are more used to the game though, quite a lot of the Timelines missions are interesting to experience.
I think there are also some youtube videos out there too with tips on how to do Timelines missions better.

and once you do the timelines scenarios and unlock all the ships and sectors, subsequent game starts will have all the ships and sectors unlocked, except the Xperimental Shuttle.
